چکیده

In order to study the kinetic behavior of ligand binding, flash photolysis is widely used with various hemoproteins [l-4]. The applicability of this method, however, seems to be limited to the ferrous ligated complexes, such as CO, O2 and NO complexes, since it has been believed that only those ferrous complexes are photodissociable. To extend this technique to the ferric state, we have examined the photodissociability of the various complexes of ferrihemoproteins.

Fourier transform infrared spectroscopy study of ligand photodissociation and migration in inducible nitric oxide synthase

Inducible nitric oxide synthase (iNOS) is a homodimeric heme enzyme that catalyzes the formation of nitric oxide (NO) from dioxygen and L-arginine (L-Arg) in a two-step process. The produced NO can either diffuse out of the heme pocket into the surroundings or it can rebind to the heme iron and inhibit enzyme action. Here we have employed Fourier transform infrared (FTIR) photolysis difference ...

Flash Photolysis Studies of the Ruthenium(II) Porphyrins Ru(P)(NO)(ONO). Multiple Pathways Involving Reactions of Intermediates with Nitric Oxide1

Described are the spectra and kinetics of transients formed by laser flash photolysis of the ruthenium nitrosyl nitrito complexes Ru(P)(NO)(ONO), P ) TPP (meso-tetraphenylporphyrin), OEP (octaethylporphyrin), TmTP (tetra(m-tolyl)porphyrin), and FTTP (tetra(m-trifluoromethylphenyl)porphyrin) in benzene solutions. Two transient decay processes are seen on the time frame (<1 ms) of the flash photo...

Kinetics, mechanism, and spectroscopy of the reversible binding of nitric oxide to aquated iron(II). An undergraduate text book reaction revisited.

A detailed kinetic and mechanistic analysis of the classical "brown-ring" reaction of [Fe(H(2)O)(6)](2+) with NO was performed using stopped-flow and laser flash photolysis techniques at ambient and high pressure. The kinetic parameters for the "on" and "off" reactions at 25 degrees C were found to be k(on) = 1.42 x 10(6) M(-1) s(-1), DeltaH(++)(on) = 37.1 +/- 0.5 kJ mol(-1), DeltaS(++)(on) = -...

Photodissociation of ligands from heme and heme proteins. Effect of temperature and organic phosphate.

The effect of temperature on ligand photodissociation from protoheme and the heme proteins hemoglobin (Hb) and myoglobin (Mb) has been examined. The quantum yield of photodissociation (phi) is greater at 40 degrees than at 0 degrees; in general, larger increases are seen in the less photosensitive complexes, while phi does not change in the most photosensitive complexes. The ratio of phi at 40 ...

Characterization of the Heme Pocket Structure and Ligand Binding Kinetics of Non-symbiotic Hemoglobins from the Model Legume Lotus japonicus

Plant hemoglobins (Hbs) are found in nodules of legumes and actinorhizal plants but also in non-symbiotic organs of monocots and dicots. Non-symbiotic Hbs (nsHbs) have been classified into two phylogenetic groups. Class 1 nsHbs show an extremely high O2 affinity and are induced by hypoxia and nitric oxide (NO), whereas class 2 nsHbs have moderate O2 affinity and are induced by cold and cytokini...
